本次講座摘要:
The process of drug discovery is still risky, lengthy, and costly. Drug repurposing offers an unconventional approach to exploring new disease indications from existing drugs. Drugs that have failed trials or have not entered them can be salvaged if novel mechanisms of action or targets are found. Polypharmacology refers to a drug’s ability to affect multiple targets, which is now accepted as a basic property of small molecules and underpins drug repurposing. Polypharmacology is useful for discovering novel mechanisms of action and improving therapeutic efficacy for different diseases. Repurposing offers cost savings and bypasses safety concerns as extensive drug data is often available. Large databases have been built in recent years and we are interested in using and integrating these for drug repurposing and polypharmacology analysis. I’ll provide an example of anticancer drug discovery and polypharmacology by integrating the Connectivity Map with other bioinformatics tools.
