5′ AMP-activated protein kinase (AMPK) is one of the central regulators of cellular metabolism that has been linked to the regulation of tumor progression and the metabolic reprogramming in hepatocellular carcinoma (HCC). In our study, we developed a novel allosteric AMPK activator, SCT-1015. Under the treatment of SCT-1015, the activated AMPK negatively regulated transcription factor HIF1α through both inducing HIF1α degradation (ERK/PHD-dependent proline hydroxylation on HIF1α) and suppressing HIF1α synthesis (mTORC1 signaling pathway). As a result of the reduced protein abundance of HIF1α, the transcription of the enzymes regulating glycolysis and oxidative phosphorylation was decreased. Such downregulation led to reduced glycolysis and increased oxidative phosphorylation thus attenuating the Warburg phenotype and eliciting an anti-HCC effect in vitro and in vivo. Our results demonstrated the role of AMPK mediated pathway in HCC metabolic reprogramming, and provided the therapeutic possibility through AMPK activation by a newly developed small molecule, SCT-1015
